Skeletal muscle plays a central role in the regulation of whole body glucose metabolism. By virtue of its mass, skeletal muscle is the primary tissue responsible for insulin-dependent glucose clearance, accounting for up to 80% of whole-body glucose uptake. Under fasting conditions, when insulin is low, muscle accounts for less than 10% of whole-body glucose uptake, as the central nervous system becomes the most important user of blood glucose. However, when circulating glucose increases, muscle becomes quantitatively the most important tissue involved in glucose metabolism.
Despite the high levels of glucose uptake that occurs in muscle, the intracellular concentration of free glucose does not change. This indicates that glucose is quickly metabolized by muscle, and that glucose transport across the cell membrane is the rate limiting step in glucose utilization...
